The Myoviridae cyanophage A-1(L) specifically infects the model cyanobacteria Anabaena sp. PCC 7120. Following our recent report on the capsid structure of A-1(L), here we present the high-resolution cryo-EM structure of its intact tail machine including the neck, tail and attached fibers. Besides the dodecameric portal, the neck contains a canonical hexamer connected to a unique pentadecamer that anchors five extended bead-chain-like neck fibers. The one-dimensional multicellular cyanobacterium Anabaena sp. PCC 7120 exhibits two different cell types under nitrogen-deprived conditions. We found that the intensity of the Raman band at 1,629 cm -1 , which is associated with phycocyanin, was higher in undifferentiated cells (vegetative cells) than in differentiated cells (heterocysts). We observed cells whose band intensity at 1,629 cm -1 was statistically lower than that of vegetative cells, and named them "proheterocysts". In the genome of the heterocystous cyanobacterium Calothrix sp. NIES-4101 (NIES-4101), the four genes essential for nitrogen fixation (nifB, nifH, nifD, and nifK) are highly fragmented into 13 parts in a 350-kb chromosomal region, and four of these parts are encoded in the reverse strand. Such a complex fragmentation feature makes it difficult to restore the intact nifBHDK genes by the excision mechanism found in the nifD gene of the Anabaena sp. PCC 7120 heterocyst.
