AIM: The "PARUS" program included investigation of the analgesic, muscle relaxant and sedative effects of Mydocalm-Richter which acts as central muscle relaxant in patients with myofascial pain syndrome, taking into account its registered indication for use - the hypertonus and cross-striated muscle spasm. MATERIALS AND METHODS: Fifty patients with myofascial trigger points, the mean age of 41.67±11.86 years, have been enrolled in the study. All patients had undergone clinical examination that allowed the diagnosis of myofascial pain syndrome...

The article deals with the dynamics of echocardioscopy indices during 1.5 years in a patient who underwent an acute myocardial infarction (MI). Two months after the MI left ventricular (LV) end-diastolic size and LV volume were 61 mm and 190 ml, respectively; LV ejection fraction (EF) was 42%. At the background of maximal possible doses of ramipril, bisoprolol, and selective antagonist of aldosterone receptors eplerenone, and after revascularization, within eight months LV end-diastolic size decreased down to 57 mm, LV volume - to 158 ml, and LVEF increased up to 51%...

The effect of mydocalm (tolperison, 5 mg/kg single dose) on the dynamics of intra-abdominal hypertension (IAH), blood circulation regulation, and oxygen balance in the tissues of intestinal wall were studied in acute experiments on rabbits. Using a special stand of original design, the initial IAH level was modeled at 200 mm H2O with the subsequent stopping of further receipt of liquid during 3 hours in an elastic container in the abdominal cavity. During 3-h observation without drug administration, no changes in IAH due to the tone of muscles of the frontal abdominal wall takes place, but there is progressive deceleration of local blood flow (-35...

An effect of mydocalm on muscle tone and functional rehabilitation in 1700 stroke patients (mean age 63,4 years) has been studied. Patients have been divided into two standardized groups: the basic group (850 persons who received mydocalm) and control one (850 persons who didn't receive mydocalm). The muscle tone was followed up using the Ashworth Scale, pain syndrome was assessed with the Huskisson visual analog scale and a multidimensional verbal-color pain test. The functional rehabilitation was evaluated using Barthel, Lindmark, Scandinavian and Merton & Sutton scales...

Tolperisone (Mydocalm) is a centrally acting muscle relaxant with few sedative side effects that is used for the treatment of chronic pain conditions. We describe three cases of suicidal tolperisone poisoning in three healthy young subjects in the years 2006, 2008 and 2009. In all cases, macroscopic and microscopic autopsy findings did not reveal the cause of death. Systematic toxicological analysis (STA) including immunological tests, screening for volatile substances and blood, urine and gastric content screening by GC-MS and HPLC-DAD demonstrated the presence of tolperisone in all cases...

A new capillary GLC method for the determination of tolperisone in human plasma was developed. Pharmacokinetic and comparative bioavailability studies were carried out after i.v. administration and after oral administration of two different preparations of tolperisone. After i.v. administration of the drug the elimination half-life was found to be 1.55 +/- 0.7 h (mean +/- S.D.), the apparent volume of distribution to be 5.1 +/- 1.0 l/kg (mean +/- S.D.) and total body clearance to be 140.8 +/- 33.8 l/h (mean +/- S...

Mydocalm was used for the treatment of 31 patients, mean age 45 +/- 4 years, with chronic headache of tension in a dosage of 150 mg 3 times per day during 4 weeks. Twenty-six patients have completed the therapy course. Before- and after treatment, neurological status, dynamics of psychoautonomic disorders, an extent of anxiety and depression, quality of sleep, quality of life, pain syndrome measured by a number of tests, were assessed, along with electromyography and examination of nociceptive reflex. Subjective evaluation of the drug efficacy was taken into account as well...

The case of anaphylactic shock due to Mydocalm administration is presented. 49-year-old woman suffered from spinal osteoarthritis and she was treated with NSAIDs and Mydocalm for many years. 6 weeks before admission to hospital the first anaphylactic shock was developed with loss of consciousness after oral administration of Mydocalm. Then she was admitted to the hospital in order to diagnose and verify the suspicion whether Mydocalm caused this reaction. Percutaneous test with Mydocalm was performed and it caused anaphylactic shock...

Four patients with anaphylaxis attributed to the intake of the centrally acting muscle relaxant tolperisone hydrochloride (Mydocalm) were observed at the Emergency Department of the Geneva University Hospital between November 2001 and March 2003. All patients were middle-aged women who took tolperisone for chronic muscular pain. All reactions occurred within an hour after oral intake of this drug frequently prescribed in Switzerland. The severity of anaphylaxis ranged from urticarial reactions to shock with arterial hypotension...

Sedative effects of single and repeated doses of 50 mg and 150 mg tolperisone hydrochloride (Mydocalm), a centrally active muscle-relaxing agent, were evaluated in a placebo-controlled double-blind clinical trial. A total of 72 healthy young adults balanced by sex were randomized to receive 50 mg or 150 mg tolperisone hydrochloride or placebo t.i.d. for a period of 8 days. Control examinations were performed in the mornings of days 1 and 8 before intake of the morning dose and at 1.5, 4 and 6 hours postdose...
