OBJECTIVE: In view of the high incidence of polycystic ovary syndrome (PCOS) and the unsatisfactory therapeutic effects of dimethyldiguanide or clomifene citrate alone, our study aimed to investigate the therapeutic effects of dimethyldiguanide combined with clomifene citrate in the treatment of PCOS. METHODS: A total of 79 patients with POCS and 35 healthy females were included, and endometrial biopsies were obtained. The sterol regulatory element-binding protein-1 (SREBP1) expression in endometrial tissues was detected by qRT-PCR...

It is established that the physiological effects of insulin are primarily mediated by the insulin signaling pathway. However, a defective insulin signaling is closely associated with the clinical manifestations of polycystic ovary syndrome (PCOS), which include excess androgen levels, insulin resistance and anovulation, and is involved in the pathophysiology of PCOS at the molecular level. Dimethyldiguanide (DMBG) has been widely employed to alleviate reproduction dysfunction in women with PCOS, however, the exact mechanism of this effect remains unclear...

Polycystic ovary syndrome (PCOS) is a major health problem in reproductive-aged women worldwide, but the precise pathogenesis of PCOS remains unclear. Our previous study revealed that hypoxia-inducible factor (HIF)-1a mediated endothelin (ET)-2 signaling plays an important role in ovulation in rats. Therefore, the present study used a PCOS rat model to test the hypotheses that HIF-1a signaling is expressed and inhibited in ovaries during PCOS formation and that the HIF-1a/ET-2 signaling pathway is a target of dimethyldiguanide (DMBG) in the clinical treatment of PCOS...

This study was designed to investigate the potential effects of 14days' intragastrically given of cinnamon polyphenols (CPS) in treating diabetic mice induced by intraperitoneal injection of streptozotocin (150mgkg(-1)) and fed high-sugar, high-fat diet. The diabetic mice model was successfully established through determining on fasting blood-glucose (FBG) test. As revealed by glucose oxidase (GOD) and radioimmunoassay (RIA), both dimethyldiguanide (DC, 0.6gkg(-1)d(-1)) and CPS (0.3, 0.6, 1.2gkg(-1)d(-1)) treatments significantly resulted in down-regulation of blood glucose and insulin levels in serum, while the levels of oxidative stress markers were markedly lowered through ELISA assay...

Early in the ancient Egyptian period 3, 500 years ago, people began to record the symptoms of diuresis related to Diabetes Mellitus. Thereafter, the cognition of Diabetes Mellitus in people evolved several times, that is: from simple symptom-recording to realizing that the essential pathogenesis is the kidney, and then recognizing that the root cause is the pancreas; from the cognition of islet cells to the separation and purification of insulin; from recognizing that sulfonamides have a hyperglycemic action to the appearance of dimethyldiguanide and the improvement of the method of Insulin's injection method...

A simple and straightforward methodology toward the synthesis of novel 4,5-biphenyl-2-pyrimidinylguanidine compounds has been developed by one-pot reaction of biguaninde or dimethyldiguanide with isoflavones. A series of 20 new compounds was reported. All of them were characterized by FT-IR, NMR, and elemental analysis. A typical compound was determined by X-ray diffraction. A variety of substrates can participate in the process with good yields and high purities, making this methodology suitable for library synthesis in drug discovery...

OBJECTIVE: To observe the therapeutic effect of Fufang Cangzhu Tang (Composite Atractylodes Decoction) on senile obesity or overweight with impaired glucose tolerance (IGT). METHODS: 32 cases of senile obesity or overweight with IGT were treated with Composite Atractylodes Decoction, with another 30 cases treated with dimethyldiguanide as the controls. Changes of body weight, waist circumference, hip circumference, waist hip circumference ratio (WHR), glucose tolerance, fast serum insulin and blood lipid before and after treatment were compared...

OBJECTIVE: To study the effect of sour TCM compound Recipe (SCCR) on insulin resistance in experimental rats with diabetes mellitus type 2 (DM2), under the guidance of TCM doctrine of "sour restrains sweet". METHODS: Model rats of DM2 were established by 8 weeks' feeding with high calorie forage combined with intraperitoneal injection of small dose of streptozotocin, and treated with SCCR (15 g/kg of crude drug/day). Levels of fasting blood glucose (FBG), serum insulin, free fatty acids (FFA), tumor necrosis factor a (TNF-alpha), combining capacity and constant of insulin receptor in liver were determined before treatment and 4, 8 and 12 weeks after treatment, and the insulin sensitive index was calculated...
