Vecuronium bromide (Piperidinium, 1-[(2β,3α,5α,16β,17β)-3,17-bis(acetyloxy)-2-(1-piperidinyl)androstan-16-yl]-1-methyl-, bromide; Norcuron®) has been extensively used in anesthesiology practice as neuromuscular blocking agent since its launch on the market in 1982. However, a detailed crystallographic and NMR analysis of its advanced synthetic intermediates is still lacking. Hence, with the aim of filling this literature gap, vecuronium bromide was prepared starting from the commercially available 3β-hydroxy-5α-androstan-17-one (epiandrosterone), implementing some modifications to a traditional synthetic procedure...

BACKGROUND: Different anesthesiological techniques are currently used for intragastric balloon positioning and removal. The aim of this study is to compare different anesthesiological approaches for balloon positioning and removal in a large multicentric patient population. METHODS: Retrospective multicenter study was conducted. From May 2000 to April 2008, 3,824 patients underwent BIB(®) placement [1,022 male/2,802 female; mean age 39.5 ± 14.7 years, range 12-71 years; mean body mass index (BMI) 44...

BACKGROUND: Varying effects of pancuronium on neonatal pulmonary mechanics have been documented, including a decrease in pulmonary compliance or no significant change in compliance; but measurements of respiratory mechanics or quantification of episodes of hypoxemia in preterm newborns receiving vecuronium (Norcuron, Bedford Labs, Bedford, OH, USA) have not been reported. The objective of the present study was to quantify the short-term effects of vecuronium on pulmonary mechanics and episodes of hypoxemia in preterm infants receiving mechanical ventilation...

Dysoxia, a state in which O2 supply is inadequate to meet tissue metabolic needs, is often first detected in regional organs such as the gut. An increase in PCO2 is believed to reflect the development of gut dysoxia. The stomach is a well-documented clinical site for detecting gut PCO2; however, measurement issues make this a less than ideal monitoring site. Other sites along the GI tract may be equally sensitive to detect changes in PCO2. Rectal CO2 measurement may have the advantage of being less invasive, low risk, and continuous without the limitations associated with gastric monitoring...

Central command and the exercise pressor reflex can independently reset the carotid baroreflex (CBR) during exercise. The present investigation assessed the interactive relationship between these two neural mechanisms in mediating baroreflex resetting during exercise. Six men performed static leg exercise at 20% maximal voluntary contraction under four conditions: control, no perturbation; neuromuscular blockade (NMB) induced by administration of the neuromuscular blocking agent Norcuron (central command activation); MAST, application of medical antishock trousers inflated to 100 mmHg (exercise pressor reflex activation); and Combo, NMB plus MAST (concomitant central command and exercise pressor reflex activation)...

Topical vecuronium bromide (Norcuron) and combinations with atropine and phenylephrine, were evaluated as mydriatics in juvenile double-crested cormorants (Phalacrocorax auritus). Nine cormorants were treated with each of four protocols: 1% atropine; 4 mg/mL vecuronium bromide (total 0.16 mg/eye); atropine with vecuronium; and atropine, 2.5% phenylephrine, followed by vecuronium. Drugs were applied topically at 15-min intervals (0.01 mL/drop). Pupil diameter was measured manually every 15 min with a pupil gauge calibrated to the nearest 0...

1. This investigation was designed to determine the contribution of central command to the resetting of the carotid baroreflex during static and dynamic exercise in humans. 2. Thirteen subjects performed 3.5 min of static one-legged exercise (20 % maximal voluntary contraction) and 7 min dynamic cycling (20 % maximal oxygen uptake) under two conditions: control (no intervention) and with partial neuromuscular blockade (to increase central command influence) using Norcuron (curare). Carotid baroreflex function was determined at rest and during steady-state exercise using a rapid neck pressure/neck suction technique...

Efficiency of ondansetron, a selective 5- HT3 receptor antagonist, in prevention of postoperative nausea and vomiting in 40 ASA I-II patients to undergo emergency intraabdominal operations is studied in a randomized, double-blind and placebo controlled study. Patients of no premedication are administered 4 mg i.v. ondansetron or placebo (saline) before induction. Thiopental (4 mg/kg) was used for induction, succinylcholine (2 mg/kg) for muscular relaxation, and 50% nitrous oxide in oxygen and isoflurane (0...

The new nondepolarizing muscle relaxant vecuronium bromide (Norcurone) was applied to 30 patients in the course of general balanced anesthesia for abdominal operations. The advantages of Norcurone over other similar nondepolarizing muscle relaxants used so far are pointed out. The fact that Norcurone has no adverse effect on the hemodynamics is considered essential advantage for its acceptance in anesthesiologic practice.

The new formula of propofol was introduced in 1985. Because of its characteristics of rapid onset and elimination, it was often used as a drug of induction and maintenance of total intravenous anesthesia. However, the hemodynamic effects of propofol was still controversial. The present study was to observe its hemodynamic reactions at higher infusion rate. Hemodynamic studies were performed in 11 female patients, 17 to 54 years old, ASA class I-II, presenting for major lower abdominal operation. Patients with heart, lung, liver, or endocrine disease were excluded...

Norcuron injection was administered before intubation in the course of anaesthesia in 57 gynaecological operations. Surgical relaxation was performed with Arduan. The combined use of the two non-depolarizing neuromuscular blocking agents proved to be favourable and useful from the anaesthesiological aspect.

Vecuronium bromide (Norcuron, Organon, Inc., West Orange, NJ) is a common neuromuscular blocking agent used to facilitate mechanical ventilation. Cases have been reported in which prolonged use of vecuronium resulted in severe motor neuropathy, with or without myopathy. However, the time course of recovery, the functional prognosis, and the use of inpatient rehabilitation is not well-established. We are reporting the functional recovery of two cases with the diagnosis of severe vecuronium motor neuropathy and/or myopathy...

A variant of a balanced anaesthesia technique with flunitrazepam, fentanyl and N2O/O2 was used. The following equipotent doses were investigated (mg/kg body weight): vecuronium 0.07/0.10; atracurium 0.35/0.50; pancuronium 0.08/0.115. Patients under neurolept, halothane or enflurane anaesthesia received Org NC 45 for muscle relaxation. Systolic, diastolic, mean arterial pressure and heart rate were monitored throughout the study. All patients showed a fall in heart rate after Org NC 45, but this was significant (p less than 0,05) only during neurolept and enflurane anaesthesia. There was no significant change in blood pressure after Org NC 45.

The neuromuscular and cardiovascular effects of vecuronium (Norcuron, ORG NC45) were studied in 40 adolescents (10-17 yr) and children (2-9 yr) anesthetized with 1.5% inspired halothane. Ten adolescents and ten children were given 20 micrograms/kg incremental doses of vecuronium to establish a cumulative dose-response curve during train-of-four stimulation. The ED95 dose was 56 micrograms/kg in children and 40 micrograms/kg in adolescents, children being significantly (P less than 0.01) more resistant to the neuromuscular effects of vecuronium than adolescents...
