This is the first study to detect the effect of calcium ions on the activity of transmembrane adenylyl cyclase (tmAC), the key enzyme of the adenylyl cyclase signaling system, under normal conditions and after a short-term exposure to exopolysaccharides (EPS) of the bacterial ring rot pathogen Clavibacter michiganensis ssp. sepedonicus (Cms). The complete nucleotide sequences of 11 wild and cultivated tomato GDP-L-galactose-phosphorylase- 1 homologs (VTC2) were identified and characterized. Using the acylation reaction with tosyl chloride of N-aminopropyl analogues of tacrine and its cyclic homologues with different size of the aliphatic cycle (5-8), we synthesized a number of new derivatives of p-toluenesulfonamide. It is shown that the synthesized hybrid compounds of tacrine and p-toluenesulfonamide are effective inhibitors of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) with the preferential inhibition of BChE. They also displace propidium from the peripheral anionic site of the electric eel AChE (Electrophorus electricus)...

The structure of the MADS-box gene HAM31 of the sunflower (Helianthus annuus) was characterized. It is shown that the product of this gene is an ortholog of the B-class MADS transcription factor PISTILLATA (Arabidopsis thaliana). Two types of transgenic tobacco plants (Nicotiana tabacum) with the constitutive expression of the HAM31 gene in the sense and antisense orientation were obtained. The effect of the deuterium concentration in the surrounding liquid medium on the appearance of open states between the nitrogen bases of a double-stranded DNA molecule was investigated. It is shown that the nature of this effect depends on the magnitude of the energy of the hydrogen bond break.

The structure and phylogeny of MADS-box genes HAM91 of sunflower (Helianthus annuus) and CDM115 of chrysanthemum (Chrysanthemum morifolium) were characterized. It is shown that these genes encode MADS-domain transcription factors, which are orthologs of TM6 (Solanum lycopersicum) and APETALA3 (Arabidopsis thaliana), respectively. We obtained two types of transgenic tobacco plants (Nicotiana tabacum) with constitutive expression of HAM91 and CDM115 genes. To model human interleukin-6 (hIL-6) associated diseases, unique mice with transgenic overexpression of human IL-6 and reporter fluorescent protein EGFP in cells of macrophage-monocyte lineage were generated using loxP-Cre system. High level of hIL-6 production by macrophages and monocytes, as confirmed in vitro in primary culture of bone marrow-derived macrophages, in vivo resulted in early postnatal death in vivo, presumably, due to the effect of overexpression of hIL-6 on hematopoiesis.

This is the first study to show that polyamine spermine, a low-molecular-weight nitrogen-containing compound, can induce autophagy in plants. This process is accompanied by an increased generation of reactive oxygen species and nitric oxide, which play a signal role and are required for triggering autophagy.

The results of the study of the effect of mononuclear dinitrosyl iron complexes (DNICs) with functional sulfur-containing ligands (NO donors) on the cell viability and metabolism of human lung fibroblasts are presented, and the efficiency of their action is evaluated. It was shown that cationic DNICs increased the cell viability of fibroblasts and demonstrated the cytoprotective properties. Fluorescent analysis revealed that the DNICs compounds decrease the mitochondrial membrane potential but do not have a significant effect on the level of glutathione and reactive oxygen species in fibroblasts...

Populations of four species of narrow-headed rats of the genus Stenocephalemys were examined for the presence of a mutation in the mitochondrial cytochrome b gene resulting in an amino acid substitution in the translated protein. This substitution was also found in hamsters of the subfamily Sigmodontinae inhabiting the highlands of the Andes. Simulation of three-dimensional structure of the protein showed that this substitution is located in the active site and may have a functional significance. We assumed an independent emergence of this substitution in the populations of two Afroalpine species of Stenocephalemys, which is the first known example of a combination of introgression and convergent molecular evolution...

In this study, we evaluated the antitumor activity of a gene therapy complex in which the tumor-specific control of the expression of the effector suicide gene FCU1 was performed using a two-vector system based on the site-specific Cre-LoxP recombinase system. The complex of interest showed a high therapeutic potential in a mouse colon adenocarcinoma model.

Seven different carotenoids with the number of conjugated double bonds (N) from 5 to 11 were incorporated in vitro into carotenoidless complexes LH2 of the sulfur bacterium Allochromatium vinosum strain MSU. The efficiency of their incorporation varied from 4 to 99%. The influence of N in the carotenoid molecules on the energy transfer efficiency from these pigments to bacteriochlorophyll (BChl) in the modified LH2 complexes was studied for the first time. The degree of genetic diversity of the steppe field mouse was estimated on the basis of the mtDNA control region variability data. Polymorphism of this mtDNA marker in S. witherbyi was more pronounced than previously assumed. The steppe field mouse population from the northern shore of Lake Manych-Gudilo includes several mitochondrial lines, which may be due to the existence of several migration flows to this area from Southwest Asia.

Arginine-containing peptides R3, R8, and R16 were obtained by solid-phase peptide synthesis, and their binding to nicotinic acetylcholine receptors (nAChRs) of muscle and neuronal (α7) types was studied by competitive radioligand assay with the use of 125 I-α-bungarotoxin. The resulting peptides exhibited a significantly greater binding activity with respect to the muscle-type nAChRs than to the α7 receptor. Thus, we have discovered a new class of nAChR ligands. Using mathematical modeling of the process of cancer treatment by the classical chemotherapy methods and therapy with biological agents, the effect of the kinetic parameters of the model on the final outcome of treatment was studied. It is established that the complete cure (i.e., the formation of a stable steady state with a zero number of cancer cells) cannot be reached by means of only classical chemotherapy.

A method for determination of hydroxylase activity of cytochrome P450 3A4 (CYP3A4) towards its substrate hydrocortisone using fluorescent analysis of the product was developed. 6β-hydroxycortisol, formed during CYP3A4-dependent electrocatalysis, has a characteristic fluorescent peak at λ = 427 ± 2 nm after treating with the sulfuric acid : ethanol (3 : 1) mixture and excitation at λ = 365 nm, which is different from the substrate (hydrocortisone) fluorescence (λ = 525 ± 2 nm). New data on the organization of genes entirely located in the open domains for chromatin transcription and occupying only one chromosome structure (interband) were obtained. The characteristic features of these genes are the small size (on average, 1-2 kb), depletion of the replicative complex proteins in the regulatory region, and the presence of specific motifs for binding transcription factors, as compared to the genes occupying two structures (interband and gray band).
