Mass spectrometry (MS) has developed over the last decades into the most informative and versatile analytical technology in molecular and structural biology (). The platform enables discovery, identification, and characterisation of non-volatile biomolecules, such as proteins, peptides, DNA, RNA, nutrients, metabolites, and lipids at both speed and scale and can elucidate their interactions and effects. The versatility, robustness, and throughput have rendered MS a major research and development platform in molecular human health and biomedical science...

The aim of this paper was to develop, validate, and utilize a sensitive liquid chromatography-tandem mass spectrometry bioanalytical method for bioequivalence/clinical trial studies conducted in human plasma. To accomplish the target, a stable labeled internal standard, that is, dexamethasone D4 was used as an internal standard to track and compensate the parent compound during processing, and extraction from plasma. The method involves a rapid liquid-liquid phase extraction from plasma, followed by reverse phase chromatography, and mass spectrometry detection, with a total run time of 3...

The anticancer drug ibrutinib was subjected to stress degradation studies under the ICH-prescribed hydrolytic, photolytic, oxidative and thermal stress conditions, and its degradation behavior was studied. A significant degradation was noted for the drug under acidic/alkaline hydrolytic, acid/alkaline photolytic, and oxidative conditions. The UPLC-UV/PDA studies revealed the generation of six degradation products ( I-VI ), and these were adequately resolved from the drug under the developed chromatographic conditions over a Kinetex® C18 (100 mm×4...

Contemporary life is mostly spent in indoor spaces like private houses, workplaces, vehicles and public facilities. Nonetheless, the air quality in these closed environments is often poor which leads to people being exposed to a vast range of toxic and hazardous compounds. Volatile organic compounds (VOCs) are among the main factors responsible for the lack of air quality in closed spaces and, in addition, some of them are particularly hazardous to the human organism. Considering this fact, we conducted daily in situ air analyses over 1 year using a gas chromatography-ion mobility spectrometry (GC-IMS) device in an indoor location...

A convenient method of applying competition experiments to devise a Hammett correlation in the dissociation by α-cleavage of 17 ionised 3- and 4-substituted benzophenones, YC6 H4 COC6 H5 [Y=F, Cl, Br, CH3 , CH3 O, NH2 , CF3 , OH, NO2 , CN and N(CH3 )2 ] is reported and discussed. The results given by this approach, which rely on the relative abundance of [M-C6 H5 ]+ and [M-C6 H4 Y]+ ions in the electron ionisation spectra of the substituted benzophenones, are compared with those obtained by previous methods...

Protonation of gabapentin was studied using corona discharge ion mobility spectrometry with two types of reactant ions at temperature of 200°C. It was found that at elevated temperatures, ionization of gabapentin proceeds via two channels, including the protonation and reactant ion attachment, in the presence of both hydronium and ammonium reactant ions. The effect of the sample concentration on the relative intensity of product ion peaks was also studied. It turned out that in high concentrations, in addition to the protonation of gabapentin, binding of the reactant ion and the formation of proton-bound dimer also occurred, while in low concentrations, the only product of the ionization process is protonated gabapentin...

The objective of this study is to gain insights into the underlying metabolic transformations that occurred during the whole progression of cecal ligation and puncture (CLP)-induced sepsis, thus providing new targets for its treatment. High-performance liquid chromatography of quadrupole time of flight mass spectrometry (HPLC-Q-TOF-MS/MS) combined with multivariate statistical techniques was used to detect the s in serum from septic mice. Fifty male mice were divided into two groups, including the sham group ( n = 7) and the CLP-induced sepsis group ( n = 43)...

Vitamin K is an essential lipophilic vitamin that acts as a coenzyme in several metabolic pathways. Accurate measurement of apolar metabolites transported by lipoproteins in serum matrices requires high-recovery extractions of vitamin K and its derivatives following standardized protocols. Conventionally developed methods in this field have predominantly employed solid-phase extraction for the measurement of vitamin K and its derivatives. In this study, our objective was to develop an enzyme-assisted extraction method for the precise measurement of vitamin K and its derivatives...

Pepsin, because of its optimal activity at low acidic pH, has gained importance in mass spectrometric proteome research as a readily available and easy-to-handle protease. Pepsin has also been study object of protein higher-order structure analyses, but questions about how to best investigate pepsin in-solution conformers still remain. We first determined dependencies of pepsin ion charge structures on solvent pH which indicated the in-solution existence of (a) natively folded pepsin (N) which by nanoESI-MS analysis gave rise to a narrow charge state distribution with an 11-fold protonated most intense ion signal, (b) unfolded pepsin (U) with a rather broad ion charge state distribution whose highest ion signal carried 25 protons, and (c) a compactly folded pepsin conformer (C) with a narrow charge structure and a 12-fold protonated ion signal in the center of its charge state envelope...

Conventional endoscopic biopsy tests are not suitable for early detection of the acute onset and progression of peptic ulcer as well as various gastric complications. This also limits its suitability for widespread population-based screening and consequently, many people with complex gastric phenotypes remain undiagnosed. Here, we demonstrate a new non-invasive methodology for accurate diagnosis and classification of various gastric disorders exploiting a pattern-recognition-based cluster analysis of a breathomics dataset generated from a simple residual gas analyzer-mass spectrometry...

Bisphenols are known endocrine disruptors commonly utilized in food packaging and storage materials, which frequently come into touch with multiple food products packed in them. The bisphenols in fish feed and other feed materials for aquatic organisms are harmful. The consumption of such marine foods is hazardous. Hence, the feed of aquatic products needs to be verified for the presence of bisphenols. The present study was focused on developing and validating a rapid, selective, and sensitive method to quantify 11 bisphenols from the fish feed with dispersive solid-phase extraction, which was cleaned by an optimized amount of activated carbon spheres and silylated by N,O-bis(trimethylsilyl)trifluoro acetamide and analyzed by gas chromatography-mass spectrometry...

Gaussian and exponentially modified Gaussian functions were incorporated into integrating algorithms used by an open-source, cross-platform tool called CycloBranch. Using our algorithm, we can separate the m/z values 694.25802 and 694.26731 (a 0.009 Da difference), where the former belongs to the most intense peak of pyoverdine D (PvdD), and the latter to the second most intense peak of pyoverdine E (PvdE) in the respective isotopic clusters of [M + Fe-H]2+ ions...

Applied sciences have increased focus on omics studies which merge data science with analytical tools. These studies often result in large amounts of data produced and the objective is to generate meaningful interpretations from them. This can sometimes mean combining and integrating different datasets through data fusion techniques. The most strategic course of action when dealing with products of unknown profile is to use exploratory approaches. For omics, this means using untargeted analytical methods and exploratory data analysis techniques...

The Fourier transform ion cyclotron resonance method holds the lead in mass accuracy and resolving power among all other mass spectrometry methods. The dynamically harmonized cell is largely responsible for the supremacy. This cell has an ideal hyperbolic trapping potential after averaging over fast cyclotron motion. Recently we have introduced an open modification of the cell (especially useful with ultrahigh magnetic fields) and have found the analytical solution for the averaged potential inside it. The voltage on specific "regularizing" electrodes determines how close a potential is to the hyperbolic one...
